Two forms of human milk bile-salt-stimulated lipase.

This study provides the first direct evidence for the existence of two active forms of the human milk enzyme bile-salt-stimulated lipase. Heparin-based affinity chromatography and size-exclusion chromatography were used to obtain two active forms of this enzyme with molecular masses of 97 and 120 kDa.

Human Milk Bile Salt-activated Lipase

Further studies on human milk bile salt-activated lipase were performed toprovide kinetic and additional chemical characterizations of this enzyme. The enzyme was homogeneous by urea-sodium dodecyl sulfatepolyacrylamide gel electrophoresis and isoelectric focusing with an isoelectric point of 3.7. A unique feature of the amino acid composition of this enzyme was a high proline content (13 mol %...

Sequence identity between human pancreatic cholesterol esterase and bile salt-stimulated milk lipase.

Three overlapping cDNA clones covering the entire primary sequence of the bile salt stimulated lipase in human milk were isolated from a human breast lambda gt10 cDNA library by screening with the rat pancreatic cholesterol esterase cDNA. Nucleotide sequencing of the cDNA showed that the human milk lipase mRNA encodes a 748-residue protein, including a 23-residue signal peptide. The human milk ...

Acyl-chain specificity of human milk bile-salt-activated lipase.

In order to probe the active-site structure of human milk bile-salt-activated lipase (BAL), the kinetics of the BAL-catalysed reaction were studied using monoesters as substrates. Among the fatty acyl chains, ranging from C8 to C16 of monoacylglycerols in a single equimolar assay mixture, there was a consistent trend of increased reactivity with decreased fatty-acyl-chain length for both the ba...

The complete digestion of human milk triacylglycerol in vitro requires gastric lipase, pancreatic colipase-dependent lipase, and bile salt-stimulated lipase.

Gastric lipase, pancreatic colipase-dependent lipase, and bile salt-stimulated lipase all have potential roles in digestion of human milk triacylglycerol. To reveal the function of each lipase, an in vitro study was carried out with purified lipases and cofactors, and with human milk as substrate. Conditions were chosen to resemble those of the physiologic environment in the gastrointestinal tr...
